The function itself tells you everything you need to know, because it's literally a one-line function:

function set_post_thumbnail_size( $width = 0, $height = 0, $crop = false ) {
     add_image_size( 'post-thumbnail', $width, $height, $crop );
}

It's as simple as that. You should use this function to set the post-thumbnail size. You should not use any other method.

The "post-thumbnail" is the size used for "Featured Images" which you enable using add_theme_support( 'post-thumbnails' ). If you have one, you should have the other as well. When in doubt, look at the default themes. Twenty-X have all had these in them.
