Yes, we are using HyperDB on WP.com and quite a few other large MU  
installs are using it as well with great success.  Since all of the  
failover is in the code, you don't have to worry about setting up,  
maintaining, and scaling another piece of the puzzle (MySQL Proxy for  
example).
